I have a pair of RTBs which I think bred. All the sudden over a 2 week period it looks like she's gotten 50%-75% bigger, so I think she's gravid. I want to handle her to keep her calm, but is it okay to handle a gravid snake?

"handle her to keep her calm" ?????????
Refrain from handling the snake if you believe she might be gravid.

I would not recommend handling a gravid Boa, unless it was absolutely necessary. It WON'T "keep her calm", trust me. If anything, it will make her more irritable. Most female Boas will try to rip you a new one when you remove their babies(which you should do as quickly as possible, to avoid her accidentally crushing any of them), no matter how gentle she was before, so that is something you must be prepared to deal with if you are going to breed these snakes. My girls would lunge and hiss like King Cobras on crack if I got anywhere near them and their babies, once they went into labor, and would stay like that for at least several days after giving birth, before they started to settle down again.
