It is hard for most people to say "I don't know."
I don't know who has the largest display.
BUT
I think you will find excellent diversity and impressive collections at both the Cape Fear Serpentarium in Wilmington, North Carolina:
http://www.capefearserpentarium.com/
and at the Kentucky Reptile Zoo in Slade, Kentucky:
http://www.geocities.com/kentuckyreptilezoo/
Hope you find what you are looking for.
Sometimes it takes more than a few minutes or hours to get a response here.

No one has mentioned the San Diego Zoo. They have a nice collection of hots from all around. mambas, cobras, eyelash vipers, bushmasters etc. One of the best collection i have seen at a zoo. But, then again i live in AZ, and the zoos out here are seriously lacking. The biggest zoo here, the Phoenix Zoo, has like 4 rattlers, some non venomous like coachwhips, gopher snakes, and a long nose, and a Burmese.

We should keep in mind that the "largest" exhibit is not necessicarily the "best".
There are many zoos with smaller collections that have some outstanding exhibits, and some very rarely seen animals.
Every zoo or museum or reptile park in the country will be different, and all will have their good and bad features.
The thing to do is see all of them, LOL !

I have been to and enjoyed the San Diego Zoo collection.
Since the question was:
"which zoo in the southeast USA has the largest reptile exhibit?"
I thought the southeast part precluded San Diego.

depending on where you are in the southeastern US, rainforest adventures in gattlinburg is probably worth the trip. i have always been impressed with the display animals there, even before they had some of mine on display. =) i would give them a try. http://www.rfadventures.com/ if you are near chattanooga, tn. i will be happy to show you a massive collection of southeastern pit vipers for free. i have over 30 horridus. probably about as many cottonmouths and maybe even more then 30 copperheads. few edb's and pygs as well. the chattanooga nature center has the largest timber rattlesnake in captivity. if you are in the area, she is worth seeing.
